miR-22 mimic is a synthetic microRNA designed to replicate the function of endogenous miR-22, which acts as a tumor suppressor in various cancers, including multiple myeloma. It works by negatively regulating the expression of DNA Ligase 3 (LIG3), a critical component of the alternative non-homologous end-joining (A-NHEJ) DNA repair pathway. By binding to the 3' UTR of LIG3 mRNA, miR-22 mimics induce its degradation or inhibit its translation, thereby suppressing A-NHEJ-driven genomic instability. This leads to cell growth inhibition, apoptosis induction, and cell cycle arrest in malignant cells. miR-22 mimics have also been shown to potentiate the cytotoxic effects of PARP inhibitors, such as olaparib, by creating a synthetic lethality effect through the simultaneous inhibition of multiple DNA repair mechanisms.
