Overview

A miR-222 inhibitor is a synthetic oligonucleotide (such as an antisense oligonucleotide, antagomir, or locked nucleic acid) designed to specifically bind to and neutralize mature microRNA-222 (miR-222). miR-222 is an oncogenic microRNA (oncomiR) that is frequently upregulated in various cancers, including breast cancer, pancreatic cancer, and multiple myeloma, where it promotes cell proliferation, migration, and drug resistance (such as adriamycin resistance) by downregulating tumor suppressors like PTEN, p27Kip1, and p57Kip2. By inhibiting miR-222, these inhibitors restore the expression of these target genes, thereby suppressing tumor growth, inducing apoptosis, and reversing chemoresistance. miR-222 inhibitors are widely used as research reagents and are being investigated in preclinical models, including exosome-based delivery systems, for their potential therapeutic applications in oncology.
