Overview

**miR-28 mimic** refers to a synthetic double-stranded oligonucleotide designed to imitate the function of the endogenous miR-28 microRNA, most often the mature "miR-28-5p" strand. miR-28 mimics restore or enhance the physiological function of miR-28, allowing exogenous modulation of target gene expression. Preclinical studies demonstrate that transfection or delivery of the miR-28 mimic suppresses expression of immune checkpoints such as PD1, TIM3, and BTLA in exhausted T cells, reinvigorates cytokine production (including IL-2 and TNF-α), and reduces traits of T cell exhaustion in models of cancer, including melanoma. In cancer cells, miR-28-5p mimics downregulate oncogenic proteins (such as RAP1B, MAD2L1), repress cell proliferation, disrupt nucleolar rRNA biogenesis by targeting the ribosomal protein mRNA RPS28, and exert tumor suppressor effects in malignancies such as renal cell carcinoma and B cell lymphoma. The mechanism relies on the RNA-induced silencing complex (RISC) pathway, where miR-28 mimic leads to target mRNA degradation or translational inhibition. As of October 2025, there is no evidence of advanced clinical development or commercial formulation of miR-28 mimic, and it is primarily used in preclinical and experimental settings[2][3][5].
