Overview

**miR-29b-chitosan** is a preclinical, systemically administered chitosan nanoparticle formulation carrying a chemically modified microRNA-29b mimic. The RNA mimic restores miR-29b activity, including repression of *LYPLA1* and broader extracellular-matrix-associated gene expression, with the intended effects of improving endothelial nitric oxide signaling and limiting pathological fibrosis. In mouse models, repeated systemic dosing reduced angiotensin II-induced hypertension and improved cardiac fibrosis, renal fibrosis, albuminuria, proteinuria, and myocardial performance in diabetic models. The published work also evaluated vascular safety in abdominal aortic aneurysm and atherosclerosis models; it reported no worsening of aneurysm size, necrotic plaque area, or mortality, although fibrotic-cap thinning was observed in an atherosclerosis model.
