miR-302 is a conserved, pluripotency-associated microRNA family (comprising miR-302a, miR-302b, miR-302c, and miR-302d) that is highly expressed in embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs). It acts as a master regulator of cell cycle progression, pluripotency, and cellular rejuvenation. miR-302 is being developed as a therapeutic agent for various indications. Notably, Prolifagen Therapeutics is developing PRO-302, a formulation of miR-302 combined with a hyaluronic acid-based hydrogel, for localized intramyocardial administration to promote cardiomyocyte proliferation and heart muscle regeneration following myocardial infarction. Additionally, miR-302 has been investigated for its potential in treating cancer (such as breast cancer, cervical cancer, and glioblastoma) by targeting cell cycle regulators (like CDK2, CDK4/6, and BMI-1) and CXCR4, as well as in Alzheimer's disease, wound healing, and bacterial pneumonia.
