miR-34a antagomir is a synthetic, chemically modified oligonucleotide designed to silence the activity of microRNA-34a (miR-34a). miR-34a is a well-characterized tumor-suppressive microRNA and a downstream transcriptional target of the p53 protein; it is frequently downregulated in various cancers, including pancreatic cancer. The antagomir works by binding with high affinity to the mature miR-34a sequence, thereby preventing it from interacting with its target mRNAs, such as Bcl-2, CDK6, SIRT1, and components of the Notch signaling pathway. In research settings, miR-34a antagomirs are primarily used as laboratory tools to investigate the functional consequences of miR-34a loss or to validate that the anti-tumor effects of certain agents (such as HDAC inhibitors or demethylating agents) are mediated through the restoration of miR-34a expression. By inhibiting miR-34a, the antagomir can rescue the expression of oncogenic targets and promote characteristics associated with cancer stem cells, such as self-renewal and epithelial-mesenchymal transition (EMT).
