miR-3662 is a microRNA located within the chromosome 6q23.3 hematopoietic quantitative trait locus (QTL). It functions as a tumor suppressor in the context of malignant hematopoiesis, particularly acute myeloid leukemia (AML). The microRNA exerts its effect by directly targeting the 3′-UTR of IKBKB (Inhibitor of nuclear factor kappa-B kinase subunit beta), which results in reduced mRNA and protein expression of IKBKB. This inhibition leads to decreased activation and nuclear localization of NF-κB, a key transcription factor in cancer cell survival. Preclinical studies have demonstrated that miR-3662 overexpression reduces cell growth and viability in AML cell lines and primary patient blasts, and significantly decreases tumor size in murine xenograft models.
