miR-377-3p antagomir is an antisense oligonucleotide therapeutic designed to specifically inhibit microRNA-377-3p (miR-377-3p) function. This synthetic oligonucleotide binds to and sequesters miR-377-3p, preventing it from binding to its target mRNAs and thereby restoring expression of genes normally suppressed by this microRNA. The antagomir is chemically modified with 2'-O-methyl and 5' cholesterol modifications to enhance stability and cellular uptake. miR-377-3p normally acts as a tumor suppressor in some contexts and an oncogene in others, depending on the tissue type and disease context. In hepatocellular carcinoma, miR-377-3p suppresses tumor growth by targeting CPT1C and inhibiting fatty acid oxidation. In lung cancer, miR-377-3p promotes tumorigenesis by targeting EGR1. The antagomir has shown therapeutic potential in multiple preclinical models including cancer and cardiovascular disease.
