miR-378a-5p mimic is a synthetic double-stranded RNA oligonucleotide designed to simulate the biological activity of the endogenous microRNA miR-378a-5p. Research presented at AACR 2019 indicates that miR-378a-5p acts as a positive regulator of melanoma progression, enhancing tumor cell invasion and vasculogenic mimicry. The mimic functions by binding to the 3' untranslated regions (UTRs) of specific target mRNAs, including SUFU, STAMBP, and KLF9, leading to their degradation or translational inhibition. This modulation results in the upregulation of pro-angiogenic factors such as VEGF and uPAR, facilitating de novo vasculogenic structure formation and endothelial cell capillary-like structure formation. While currently utilized primarily as a research tool to elucidate the molecular mechanisms of melanoma metastasis and angiogenesis, miR-378a-5p and its downstream pathways represent potential targets for therapeutic intervention.
