Drug intelligence / Profile preview
miR-519c-5p inhibitor is an antisense oligonucleotide or antagomir designed to target and inhibit microRNA-519c-5p (miR-519c-5p). In preclinical models of sepsis-associated acute kidney injury (SA-AKI), miR-519c-5p is delivered to renal macrophages via neutrophil-derived exosomes (TNF-Exo), where it suppresses the expression of tumor necrosis factor alpha-induced protein 3 (TNFAIP3/A20). This suppression enhances NEK7 K63-linked ubiquitination and activates the NLRP3 inflammasome, leading to macrophage pyroptosis and severe renal injury. Treatment with a miR-519c-5p inhibitor blocks this pathway, reducing macrophage pyroptosis, alleviating renal injury, and improving survival in sepsis mouse models.
