Overview

miR-567 is a microRNA that functions as a tumor suppressor in melanoma. Research indicates that miR-567 expression is significantly reduced in melanoma tissues compared to benign nevi, and its downregulation is associated with poorer patient survival. As a therapeutic candidate, miR-567 acts by directly targeting and silencing the mRNA of key oncogenic drivers, including Insulin-like Growth Factor 1 Receptor (IGF1R), E2F Transcription Factor 1 (E2F1), and Cyclin B2 (CCNB2). This multi-targeted inhibition results in the suppression of the MAPK/ERK and PI3K/AKT signaling pathways, leading to reduced tumor cell proliferation, migration, and invasion, even in BRAF-inhibitor resistant cells. Furthermore, miR-567 has been shown to modulate the tumor microenvironment by inhibiting the growth-promoting effects of M2-polarized and melanoma-associated macrophages (MAMs).
