miR-579-3p is a tumor-suppressive microRNA that acts as a master regulator of melanoma progression and drug resistance. It functions by post-transcriptionally downregulating the expression of key oncoproteins, specifically BRAF and MDM2. Research indicates that miR-579-3p levels are high in nevi but significantly decline in stage III-IV melanomas and further decrease in cells that have developed resistance to BRAF and MEK inhibitors. Therapeutic restoration or overexpression of miR-579-3p has been shown to inhibit melanoma cell growth and migration, induce apoptosis, and impair the establishment of resistance to targeted therapies. It is being investigated as a potential therapeutic intervention for BRAF-mutated metastatic melanoma.
