miR-616 inhibitor refers to a class of experimental RNA-based agents, such as antisense oligonucleotides or shRNA, designed to antagonize the function of microRNA-616 (miR-616). miR-616 has been identified as an oncomiR that is significantly overexpressed in androgen-independent (castration-resistant) prostate cancer compared to androgen-dependent stages and benign tissues. It promotes tumor progression by directly targeting and suppressing the expression of the tumor suppressor Tissue Factor Pathway Inhibitor-2 (TFPI-2), which in turn stimulates cell proliferation and migration. Preclinical research, including studies using lentiviral-mediated knockdown, has demonstrated that inhibiting miR-616 can restore TFPI-2 levels, diminish cancer cell proliferation in vitro, and repress tumor growth in vivo. These findings suggest that miR-616 is a potential therapeutic target for preventing or treating the progression of prostate cancer to its more aggressive, treatment-resistant forms.
