miR-634 is a microRNA that acts as a tumor suppressor by post-transcriptionally regulating multiple oncogenic pathways. In the context of glioblastoma, miR-634 expression is up-regulated by the PPARα activator fenofibrate. It has been shown to directly target the 3'-UTR of CYR61 (Cysteine-rich angiogenic inducer 61), thereby inhibiting cancer cell proliferation. Additionally, miR-634 targets TSC2 (Tuberous Sclerosis Complex 2), a negative regulator of the mTOR signaling pathway, leading to increased phosphorylation of p70 S6 kinase. Beyond glioblastoma, miR-634 has been investigated for its ability to sensitize various cancer cells to chemotherapy by targeting genes involved in mitochondrial homeostasis, anti-apoptosis, and drug efflux. Therapeutic strategies involving miR-634 typically utilize miR-634 mimics to restore its tumor-suppressive function in malignant cells.
