miR-654 mimic refers to a synthetic, exogenously delivered oligonucleotide that closely resembles the mature form of the naturally occurring microRNA miR-654 (either miR-654-3p or miR-654-5p) and is used to upregulate or “mimic” its activity in cells or organisms. It is typically used as an experimental therapeutic or research tool. miR-654 mimics exert their effect by increasing the intracellular levels of the respective miR-654 strand, thus promoting downregulation of target mRNAs through RNA-induced silencing complex (RISC) activity. Studies indicate that overexpression of miR-654-3p or miR-654-5p via mimics can suppress tumor growth, proliferation, migration, and invasion in a variety of cancer models, and influence chemosensitivity and epithelial-to-mesenchymal transition. For example, in non-small cell lung cancer, the miR-654-3p mimic decreased PLK4 mRNA and protein levels, leading to reduced tumor growth[1]. In colorectal cancer, miR-654-3p mimic downregulated SRC and inhibited invasion and metastasis[2]. In other models, distinct targets like CREB1, PSEN1, or QPRT have been validated, mediating tumor-suppressor or context-dependent oncogenic effects[3][5]. miR-654-5p mimics have also shown utility as experimental tools for modulating cell migration and proliferation in vascular disease or cancer[7].
