Overview

MIR002 is a novel retinoid-related molecule (RRM) that acts as an inhibitor of DNA polymerase alpha 1 (POLA1). It belongs to a class of compounds, including CD437 and ST1926, that exert anti-proliferative and pro-apoptotic effects independent of retinoid receptor-mediated transcriptional activity. MIR002 has demonstrated potent antitumor activity in preclinical models of malignant mesothelioma and non-small cell lung cancer (NSCLC), including models resistant to other RRMs due to POLA1 mutations (e.g., L764F). Mechanistically, it induces G1/S phase arrest and activates pro-apoptotic pathways. Preclinical data suggests it is not a substrate for P-glycoprotein (Pgp), indicating potential for oral bioavailability.
