MIR075 is a novel retinoid-related molecule (RRM) that acts as an inhibitor of DNA polymerase alpha 1 (POLA1). Developed through a collaboration involving Biogem and the University of Milan, it belongs to a class of compounds (including CD437 and ST1926) that exert anti-proliferative and pro-apoptotic effects independent of retinoid receptor-mediated transcriptional activity. Preclinical studies have demonstrated that MIR075 induces significant tumor growth inhibition in glioblastoma models, specifically showing a 72% reduction in tumor growth in intracranial U-87MG cell models. Unlike some other RRMs, MIR075 and its analogs are not substrates for P-glycoprotein (Pgp), suggesting potential for oral bioavailability.
