Drug intelligence / Profile preview
The miR433-3p tough decoy inhibitor (miR-433-3p TuD) is a preclinical research tool designed to competitively inhibit and sequester the microRNA miR-433-3p. Custom-synthesized as a synthetic RNA construct with tandem miR-433-3p binding sites, the decoy features a secondary structure that resists degradation. It is typically delivered via lentiviral or transgenic expression (such as knock-in at the Rosa26 locus) to study the role of miR-433-3p in bone formation, osteoblast differentiation, and growth plate dynamics. By sequestering miR-433-3p, the decoy prevents the microRNA from binding and suppressing its endogenous mRNA targets, which include Runx2, Hsd11b1, Sost, Dkk1, Hdac6, and Hif1a. In animal models, suppression of miR-433-3p activity in the osteoblast or osteochondral lineage increases trabecular bone volume, enhances bone formation, and blunts glucocorticoid-induced bone loss, though it can also disrupt growth plate development.
