miR4458HG siRNA is an experimental small interfering RNA designed to silence the long non-coding RNA (lncRNA) miR4458HG. In hepatocellular carcinoma (HCC), miR4458HG acts as an oncogene by binding to the m6A reader IGF2BP2, which stabilizes the transcripts of glycolytic enzymes such as hexokinase 2 (HK2) and glucose transporter 1 (GLUT1/SLC2A1). This activation of the glycolysis pathway fuels tumor cell proliferation and colony formation. Additionally, miR4458HG is secreted in exosomes to promote the M2-like polarization of tumor-associated macrophages by increasing ARG1 expression. By targeting miR4458HG, this siRNA inhibits these metabolic and microenvironmental pro-tumorigenic signals, suggesting a potential therapeutic strategy for HCC patients with high glucose metabolism.
