miRNA-375-3p inhibitor is a synthetic antisense oligonucleotide, specifically utilizing Locked Nucleic Acid (LNA) technology, designed to target and downregulate microRNA-375-3p. Research presented at the American Heart Association (AHA) 2025 meeting by investigators from University Hospital Halle demonstrates that miR-375-3p is significantly upregulated in senescent human coronary artery endothelial cells (HCAEC) and aged murine aortic tissues. The inhibitor aims to restore vascular function by improving angiogenic capacities, such as tube formation and sprouting, which are typically impaired during cellular aging. However, its effects on senescence are complex, as inhibition has been associated with increased cell death in non-senescent cells and the induction of pro-inflammatory cytokines like TNF-alpha and IL-1beta in senescent cells, suggesting its therapeutic application requires further refinement.
