Overview

AR1005 is a fixed-dose combination (FDC) drug candidate being developed by AriBio for the treatment of Lewy body disease (LBD), including dementia with Lewy bodies. The product combines mirodenafil (also known as AR1001), a selective phosphodiesterase 5 (PDE5) inhibitor, with rivastigmine, a dual acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitor that is a standard-of-care treatment for LBD. Mirodenafil is intended to enhance the nitric oxide (NO)/cGMP signaling pathway, promoting neuroprotection, synaptic plasticity, and improved cerebral blood flow, while rivastigmine addresses the cholinergic deficit characteristic of the disease. By combining these two mechanisms, AR1005 aims to provide synergistic improvements in cognitive function, behavioral psychological symptoms, and motor fluctuations. The drug is currently being evaluated in a Phase 2a clinical trial (NCT06537076) in patients with cognitive impairment due to Lewy body disease.
