Overview

Mirvetuximab soravtansine is a first-in-class antibody-drug conjugate (ADC) composed of a chimeric IgG1 monoclonal antibody (M9346A) targeting folate receptor alpha (FRα), which is overexpressed in certain epithelial tumors. The antibody is covalently linked via a cleavable disulfide linker to the cytotoxic agent DM4 (soravtansine), a maytansinoid microtubule inhibitor. Upon binding to FRα on tumor cells, the ADC-receptor complex is internalized and DM4 is released intracellularly, leading to cell-cycle arrest and apoptosis. Mirvetuximab soravtansine was developed primarily for the treatment of adult patients with FRα-positive, platinum-resistant epithelial ovarian, fallopian tube, or primary peritoneal cancer who have received one to three prior systemic therapies[3][4][5][9].
