MITO-66 is a novel and specific small molecule inhibitor of the mitochondrial pyruvate carrier (MPC) developed by MPC Therapeutics. It is primarily utilized as a research reagent (RUO) for the ex vivo metabolic reprogramming of T cells during the manufacturing of cell therapies, such as CAR T cells. By inhibiting the MPC, MITO-66 restricts the entry of pyruvate into the mitochondria, shifting cellular metabolism toward pathways that favor the induction of a stem cell-like memory T cell (TSCM) phenotype. This metabolic shift has been shown in preclinical studies to significantly enhance the antitumor efficacy, persistence, and fitness of CAR T cells, particularly in the treatment of human B-cell acute lymphoblastic leukemia (B-ALL). MITO-66 serves as a more specific and potentially safer alternative to earlier MPC inhibitors like UK-5099 for clinical manufacturing applications.
