Mito-CD147 single-chain variable fragment (mito-CD147 scFv) is an intracellularly expressed biologic therapeutic that specifically targets mitochondrial CD147 (Basigin). This single-chain variable fragment disrupts the mitochondrial CD147/MCT1/LDHB complex, which is involved in lactate uptake and oxidation in metastatic breast cancer cells. By blocking this complex, the compound inhibits lactate-induced stemness potential in cancer cells, depletes circulating breast cancer cells, and reduces metastatic burden. The therapeutic was demonstrated in preclinical studies to effectively inhibit metastasis in breast cancer models.
