Overview

Mito-EV is an experimental exosome-based therapy consisting of extracellular vesicles (EVs) that encapsulate stressed or damaged mitochondria. Originally identified as being released by adipocytes during thermogenesis or under conditions of obesity-induced stress, these vesicles facilitate the intercellular transfer of mitochondrial content to recipient cells. In the context of oncology, specifically glioblastoma (GBM), Mito-EVs are utilized to deliver stressed mitochondria into tumor cells, where they integrate into the existing mitochondrial network and induce significant oxidative stress. This metabolic disruption sensitizes the GBM cells to ferroptosis, an iron-dependent pathway of regulated cell death characterized by lipid peroxidation. Preclinical studies indicate that Mito-EVs can effectively reduce tumor growth and enhance the efficacy of ferroptosis activators, suggesting a novel therapeutic strategy for aggressive and treatment-resistant brain tumors.
