Drug intelligence / Profile preview
Mitoapocynin is a mitochondria-targeted synthetic analog of apocynin developed as a therapeutic antioxidant and neuroprotective agent. It is synthesized by conjugating the apocynin moiety to a lipophilic triphenylphosphonium (TPP+) cation via an alkyl chain (typically 2 or 11 carbons, referred to as C2 or C11), which facilitates its selective accumulation within the mitochondrial matrix. Mitoapocynin acts primarily as an inhibitor of NADPH oxidase (specifically NOX2 and NOX4), thereby reducing the production of reactive oxygen species (ROS) and mitigating oxidative stress, neuroinflammation, and mitochondrial dysfunction. It has been investigated in preclinical models for neurodegenerative and neurotoxic conditions, including Parkinson's disease, Alzheimer's disease, organic dust exposure-induced neurotoxicity, and organophosphate-induced status epilepticus (epilepsy).
