Overview

Mitocurcumin is a **mitochondria-targeted derivative of curcumin**, created by conjugating curcumin to a triphenylphosphonium group. This modification enhances mitochondrial localization and biological activity. It demonstrates significantly greater **antitumor** and **antibacterial** efficacy than unmodified curcumin. Mechanistically, mitocurcumin accumulates within mitochondria, induces substantial **reactive oxygen species (ROS) production**, leads to mitochondrial dysfunction, DNA damage, and triggers **intrinsic apoptotic pathways** in cancer cells. In infectious applications, mitocurcumin exhibits bactericidal activity, particularly against methicillin-resistant *Staphylococcus aureus* (MRSA), via a redox mechanism that is not affected by typical antibacterial resistance factors. Preclinical evidence suggests activity in various models of drug-resistant cancers (e.g., non-small cell lung cancer, acute myeloid leukemia), and as an antibacterial, with no current clinical approval status[1][2][3][4][7].
