Mitoxantrone + cyclophosphamide + vincristine is a multi-agent chemotherapy regimen, sometimes referred to as CMV, used primarily as antineoplastic therapy for hematological malignancies and certain solid tumors. All three agents are cytotoxic small molecules with distinct but complementary mechanisms of action: - **Mitoxantrone** is an anthracenedione analog that intercalates into DNA and inhibits topoisomerase II, leading to DNA strand breaks and inhibition of DNA replication and repair. - **Cyclophosphamide** is an alkylating agent that cross-links DNA strands, impairing cell division and leading to cell death. - **Vincristine** is a vinca alkaloid that binds to tubulin, inhibiting microtubule formation and thus arresting cells in metaphase. This combination has been used investigationally and clinically for diseases such as non-Hodgkin lymphoma and advanced breast cancer, especially as an alternative to anthracycline-containing regimens when cardiotoxicity is a concern[3][9][1]. Primary indications have included lymphoma (notably non-Hodgkin lymphoma), certain leukemias, and metastatic breast cancer, particularly in patients for whom doxorubicin is contraindicated[1][3][9]. All three drugs are administered intravenously in cyclic regimens under specialist supervision.
