Mitoxantrone + interferon beta-1a is a sequential combination therapy regimen investigated for the treatment of early-stage multiple sclerosis (MS) in patients with a high risk of disease progression. The regimen consists of an induction phase with mitoxantrone, a synthetic anthracenedione antineoplastic agent that functions as a DNA intercalator and DNA topoisomerase 2 inhibitor, providing rapid and potent immunosuppression. This is followed by a maintenance phase with interferon beta-1a (specifically the brand Rebif), a recombinant protein biologic that acts as an agonist at the interferon alpha/beta receptor to provide long-term immunomodulatory effects. This sequential approach, evaluated in the Phase 3 MITOX-REBIF trial sponsored by Rennes University Hospital, is designed to capitalize on the immediate disease-stabilizing effects of mitoxantrone and the sustained therapeutic benefits of interferon beta-1a to synergistically control disease activity and prevent disability progression.
