Overview

Mitoxantrone + paclitaxel is a combination chemotherapy regimen consisting of two small molecule antineoplastic agents, mitoxantrone and paclitaxel. Mitoxantrone is an anthracenedione derivative that intercalates into DNA, causing crosslinks and strand breaks, and inhibits topoisomerase II, leading to impaired DNA replication and repair. Paclitaxel is a taxane that stabilizes microtubules by promoting their assembly and preventing depolymerization, thereby inhibiting cell division during mitosis. This combination has been studied primarily in the treatment of metastatic breast cancer as salvage therapy after prior chemotherapy failure[1][6]. The regimen has demonstrated objective response rates with manageable toxicity profiles but carries risks such as neutropenia and cardiotoxicity[1]. Both drugs are also used individually for other malignancies.
