Drug intelligence / Profile preview
Mitoxantrone hydrochloride liposome is a pegylated liposomal nanoformulation of the synthetic anthracenedione antineoplastic agent mitoxantrone. Developed by CSPC Pharmaceutical Group, this formulation is designed to improve the therapeutic index of conventional mitoxantrone by utilizing a pegylated liposomal delivery system. This system enhances drug stability, prolongs systemic circulation, and promotes preferential accumulation in tumor tissues through the enhanced permeability and retention (EPR) effect. Mechanistically, mitoxantrone acts as a type II topoisomerase inhibitor and DNA intercalator, leading to DNA strand breaks and the inhibition of nucleic acid synthesis. The liposomal encapsulation significantly reduces the distribution of the drug to healthy tissues, thereby mitigating dose-limiting toxicities such as cardiotoxicity and severe myelosuppression. It was first approved in China in 2022 for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL).
