Drug intelligence / Profile preview
Mitoxantrone hydrochloride liposome (PLM60) is a liposomal formulation of the small molecule anthracenedione anti-tumor drug mitoxantrone. Developed by CSPC Pharmaceutical Group, it utilizes a liposomal delivery system to exploit the enhanced permeability and retention (EPR) effect, allowing for passive targeting of tumor tissues. This formulation significantly improves the pharmacokinetic profile by prolonging the drug's half-life and increasing concentration within the tumor while reducing systemic cardiotoxicity compared to conventional mitoxantrone. It acts primarily as a DNA intercalator and an inhibitor of topoisomerase II, leading to DNA strand breaks and inhibition of DNA synthesis. In China, it is approved under the brand name Duoenda for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL) and is being investigated in various regimens, such as CMOP±R, for newly diagnosed non-Hodgkin lymphoma.
