Drug intelligence / Profile preview
Duonda (mitoxantrone hydrochloride liposome) is a liposomal formulation of the anthracenedione antineoplastic agent mitoxantrone, developed by CSPC Pharmaceutical Group. It acts as a DNA intercalator and a topoisomerase II inhibitor, leading to DNA strand breaks and inhibition of DNA synthesis. The liposomal encapsulation is designed to improve the pharmacokinetic profile of mitoxantrone, enhancing tumor accumulation via the enhanced permeability and retention (EPR) effect while reducing systemic toxicities, particularly cardiotoxicity, compared to conventional mitoxantrone. It was first approved in China in January 2022 for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL) and is currently being investigated for other hematologic malignancies, including diffuse large B-cell lymphoma (DLBCL).
