Mitoxantrone hydrochloride liposome + PD-1 inhibitor is a combination chemo-immunotherapy regimen being investigated for the treatment of recurrent or metastatic nasopharyngeal carcinoma (NPC). The regimen consists of a liposomal formulation of mitoxantrone, a synthetic anthracenedione that inhibits topoisomerase II and intercalates into DNA to induce cell death, and a PD-1 inhibitor (such as tislelizumab, camrelizumab, or toripalimab) which blocks the programmed cell death protein 1 receptor to restore anti-tumor T-cell activity. This specific combination is being evaluated in a Phase 2 clinical trial (NCT05482243) led by Ming-Yuan Chen at the Sun Yat-sen University Cancer Center, specifically targeting patients who have progressed after standard platinum-based chemotherapy and/or prior immunotherapy. The liposomal delivery of mitoxantrone is designed to improve the drug's pharmacokinetic profile and reduce cardiotoxicity compared to the conventional formulation.
