Drug intelligence / Profile preview
Mitoxantrone-liposome is a **liposomal formulation of mitoxantrone**, a synthetic anthraquinone antineoplastic agent. Mitoxantrone acts by **intercalating into DNA, inhibiting topoisomerase II, disrupting nucleic acid synthesis, and inducing DNA strand breaks**, leading to cell death. The **liposomal encapsulation** enhances the pharmacokinetics and biodistribution, prolongs circulation time, allows higher accumulation in tumor tissues, and is associated with reduced toxicity, especially lower cardiotoxicity compared to conventional mitoxantrone. This formulation is primarily investigated and approved in China for the **treatment of relapsed or refractory peripheral T-cell lymphoma**, and it is being studied for uses in other solid tumors, including breast cancer, ovarian cancer, and head and neck squamous cell carcinoma. The most studied product in literature is PLM60. The liposome-modified delivery is not approved in the US for any indication as of July 2025[1][4][5][7][9].
