Mitozolomide is an imidazotetrazine derivative and an alkylating agent that was developed as an experimental antineoplastic (anticancer) drug. It acts via methylation of DNA, leading to DNA damage and cell death through apoptosis, in a mechanism similar to related agents like temozolomide. Mitozolomide was evaluated in the 1980s and 1990s but was never approved for clinical use due to high toxicity and lack of efficacy compared to successors. Its major activity involves methyl group transfer to guanine residues in DNA, resulting in cytotoxicity in rapidly dividing tumor cells.
