Overview

Mitsugumin-53 (MG53), also known as TRIM72 or tripartite motif-containing protein 72, is a member of the tripartite motif (TRIM) family of proteins. It functions primarily as a membrane repair protein in striated muscle and other tissues. MG53 acts by recruiting essential repair components such as dysferlin and serving as a scaffold to facilitate rapid cell membrane resealing after injury. Mechanistically, it operates both through its E3 ubiquitin ligase activity—targeting proteins like insulin receptor substrate 1 (IRS-1) and focal adhesion kinase for ubiquitination—and by directly participating in the assembly of membrane repair complexes at sites of damage. Recombinant human MG53 (rhMG53) is being developed as a therapeutic agent for conditions involving acute tissue injury and impaired membrane repair such as myocardial infarction, muscular dystrophy, acute kidney injury, and acute lung injury. Preclinical studies have shown that administration of rhMG53 can reduce tissue damage following ischemia-reperfusion events in animal models[1][3][5][7].
