MIV-160 is a small molecule nonnucleoside reverse transcriptase inhibitor (NNRTI) belonging to the urea phenylethylthioureathiazole (PETT) family. It is being developed as a topical microbicide for the prevention of HIV transmission. MIV-160 is a next-generation analog of MIV-150, designed with improved aqueous solubility, reduced hydrophobicity, and a more efficient synthetic pathway. It exhibits potent antiviral activity against HIV-1, including the common K103N resistance mutant, by binding to and inhibiting the HIV reverse transcriptase enzyme. The drug has been evaluated in preclinical models using delivery systems such as intravaginal rings and carrageenan-based gels, demonstrating significant protection against vaginal SHIV-RT infection in macaque studies.
