Overview

Mivavotinib is an investigational, orally administered small molecule inhibitor that targets spleen tyrosine kinase (SYK). It is being developed for the treatment of relapsed or refractory non-germinal center B-cell-like (non-GCB) diffuse large B-cell lymphoma (DLBCL), particularly in patients with activated B-cell-like disease and those harboring MYD88 and/or CD79b mutations. Preclinical studies have shown enhanced SYK activity and greater sensitivity to SYK inhibition in DLBCL tumor cell lines with these mutations. In prior phase 1/2 studies, mivavotinib demonstrated a higher response rate in non-GCB DLBCL compared to GCB DLBCL. The drug is currently under investigation as a monotherapy in phase 2 clinical trials for this patient population[6][8].
