Overview

Mivebresib (ABBV-075) is an orally bioavailable, small-molecule pan-inhibitor of the bromodomain and extra-terminal (BET) family of proteins, including BRD2, BRD4, and BRDT. Developed by AbbVie, it functions as an epigenetic modulator by binding to the bromodomains of BET proteins, which are "readers" of acetylated lysine residues on histone tails. By preventing BET proteins from binding to chromatin, mivebresib inhibits the transcription of key oncogenic drivers such as MYC. It has been investigated in Phase 1 clinical trials for the treatment of advanced solid tumors (including breast, prostate, and lung cancers) and hematologic malignancies (such as acute myeloid leukemia and multiple myeloma), both as a monotherapy and in combination with other agents like venetoclax.
