Overview

**mivebresib + venetoclax** is an investigational combination therapy for relapsed/refractory acute myeloid leukemia (AML). Mivebresib (ABBV-075) is a potent, oral, small-molecule pan-bromodomain and extraterminal (BET) inhibitor that targets BET family proteins, epigenetic readers involved in the regulation of oncogenic transcriptional programs[1][2][4]. Venetoclax is a selective small-molecule inhibitor of the antiapoptotic protein BCL-2, which disrupts leukemic stem cell survival and causes apoptosis in cancer cells[2][4]. The combination exploits the synergistic antileukemic activity: BET inhibition disrupts cancer-related gene expression and enhances cell sensitivity to apoptosis, while BCL-2 inhibition directly induces programmed cell death. This dual mechanism is being evaluated based on its preclinical and early clinical antitumor effects in AML[1][2][4][5].
