Mixed amphetamine salts is a central nervous system (CNS) stimulant medication primarily used for the treatment of attention-deficit hyperactivity disorder (ADHD) and narcolepsy. It consists of a specific 3:1 ratio of dextroamphetamine and levoamphetamine salts, specifically dextroamphetamine saccharate, amphetamine aspartate monohydrate, dextroamphetamine sulfate, and amphetamine sulfate. The drug works by increasing the synaptic concentrations of the catecholamines dopamine and norepinephrine. This is achieved through several mechanisms: the inhibition of reuptake transporters (DAT and NET), the promotion of neurotransmitter release from presynaptic neurons via interaction with the trace amine-associated receptor 1 (TAAR1), and the modulation of the vesicular monoamine transporter 2 (VMAT2). Originally developed by Richwood Pharmaceuticals (later acquired by Shire, now Takeda), it is available in both immediate-release and extended-release formulations.
