Mixed bacteria vaccine (MBV), also known as Coley's toxins, is a biological response modifier composed of a pyrogenic lysate derived from killed Streptococcus pyogenes and Serratia marcescens. Developed in the late 19th century by William Coley, it was originally used as an experimental immunotherapy for cancer. The active components are believed to include lipopolysaccharide (LPS) from the Gram-negative Serratia cell wall and streptokinase from Streptococcus. MBV stimulates immune responses, including production of interleukin-2 (IL-2), interferon (IFN), and tumor necrosis factor (TNF). Historically, it was administered via injection for cancer treatment and prevention of respiratory infections but has largely fallen out of use due to lack of proven efficacy and potential risks. More recently, oral or sublingual forms have been explored for preventing recurrent respiratory tract infections and asthma exacerbations[1][3][6].
