**MJ7-Ngb** is a preclinical, mouse-targeted anti-vascular immunotoxin comprising the rat anti-mouse endoglin monoclonal antibody clone MJ7/18 covalently linked through an SPDP linker to nigrin b, a type 2 ribosome-inactivating protein from *Sambucus nigra*. The MJ7/18 antibody directs the construct to mouse endoglin, also known as CD105, expressed on tumor neovasculature and on CD105-positive murine melanoma cells. After target-cell binding and internalization, nigrin b catalytically inactivates eukaryotic ribosomes by depurinating 28S ribosomal RNA, thereby blocking protein synthesis and inducing cytotoxicity. In a B16MEL4A5 mouse melanoma model, MJ7-Ngb inhibited early tumor growth, whereas the unconjugated antibody and nigrin b alone did not show comparable antitumor activity.
