MK-0472 is an investigational small molecule inhibitor of Src homology phosphotyrosyl phosphatase 2 (SHP2, also known as PTPN11), being developed by Merck Sharp & Dohme for the treatment of advanced or metastatic solid tumors. SHP2 is a protein tyrosine phosphatase involved in cell signaling pathways that regulate cell growth and survival, and its inhibition may disrupt oncogenic signaling in cancer cells. MK-0472 is currently being evaluated as monotherapy and in combination with pembrolizumab (a PD-1 inhibitor) or MK-1084 in phase 1 clinical trials for patients with histologically or cytologically confirmed advanced/metastatic solid tumors[1][4][5][7].
