Overview

MK-0493 is a novel, potent, and selective small molecule agonist of the melanocortin receptor 4 (MC4R), a G protein-coupled receptor involved in the regulation of food intake and energy homeostasis. Developed by Merck & Co, it was investigated as an anti-obesity agent based on strong genetic and preclinical evidence implicating MC4R in body weight regulation. In clinical trials, MK-0493 demonstrated only modest reductions in energy intake and body weight that were not statistically significant compared to placebo after 12–18 weeks of treatment. As a result, development for obesity was discontinued after phase II trials[1][2][4][6][7][8].
