MK-0668

MK-0668 is a potent, orally active small molecule antagonist of very late antigen-4 (VLA-4), also known as integrin alpha-4/beta-1. It was developed by Merck & Co. for potential use in immune system and digestive system disorders, with Crohn's disease noted as an inactive indication. The compound exhibits extremely high potency against the unactivated form of VLA-4, with picomolar activity in human whole blood and slow dissociation rates, which may help overcome poor pharmacokinetic profiles typical of this class. Its mechanism involves blocking the interaction between VLA-4 and its ligands, thereby modulating leukocyte adhesion and migration implicated in inflammatory diseases[3][4]. The drug has been studied preclinically but has not advanced to clinical approval.
