MK-1029 is an oral small molecule drug developed by Merck Sharp & Dohme for the treatment of asthma. It acts as a potent and specific antagonist of the Chemoattractant Receptor-homologous molecule expressed on Th2 cells (CRTH2), also known as prostaglandin D2 receptor 2 (DP2). CRTH2 is implicated in the pathogenesis of asthma through its role in mediating inflammation driven by Th2 cells. By inhibiting CRTH2, MK-1029 was intended to reduce airway inflammation and improve lung function in patients with persistent or uncontrolled asthma, including those not adequately managed with standard therapies such as montelukast. Clinical development included multiple phase II trials assessing efficacy, safety, and tolerability both as monotherapy and in combination with montelukast. Development has since been discontinued for all indications[3][5][2].
