MK-1169 (also known as MDK-1169) is a discontinued, preclinical-stage synthetic peptide agonist of the interleukin-2/15 receptor beta-gamma complex (IL-2/15Rβγc) developed by Medikine. Discovered using Medikine's PEPTIKINE™ platform, MK-1169 is structurally unrelated to native IL-2 or IL-15 but binds to the IL-2Rβ and IL-2Rγc subunits to induce receptor signaling. By selectively targeting the intermediate-affinity IL-2Rβγc complex and avoiding interaction with the IL-2Rα subunit (CD25), MK-1169 is designed to stimulate effector T cells and natural killer (NK) cells without activating immunosuppressive regulatory T cells (Tregs). The compound was developed for the treatment of neoplasms (cancer), and its technology served as the foundation for next-generation candidates like the Fc-fused peptide agonist MDK-202.
