Overview
MK-1248 is a humanized immunoglobulin G4 (IgG4) monoclonal antibody agonist that targets the glucocorticoid-induced tumor necrosis factor receptor (GITR). GITR is expressed on regulatory T cells (Treg), resting CD4+ and CD8+ T cells, natural killer (NK) cells, and NKT cells. Activation of GITR by MK-1248 decreases Treg-mediated suppression and enhances proliferation, effector function, and survival of T cells. The drug was developed for use in cancer immunotherapy, particularly for advanced solid tumors. In clinical studies, it has been evaluated both as monotherapy and in combination with pembrolizumab. The primary developer is Merck & Co[1][2][3][6].
